A Study on the Activities of Adenosine Deaminase Isozymes in the Red Blood Cells of normal and Phenylhydrazine

Abstract
In order to establish a general pattern of synthesis and break down of cytosolic and mitochondrial adenosine deaminase (Adenosine aminodehydrolase, EC 3.5.4.4) isozymes in nomal and reticuloeytic red blood cells of domestic rabbits, sequential measurements of total and ralative activities of isozymes in the red blood cells of normal andphenylhydraine-treated rabbits were conducted in the present study.
The results were summa rized as follows :
1. The reticuloeyte count of normal rabbit blood was approximately 2.5% and the administration of phenydrazine resulted in rapid and profound reticulocytosis, i.e. about 75% on 5th day and essentially 100% on 7th day.
2. Total activity of adenosine deaminase in normal red blood cells was 211 units and the relative activities of cytosolic and mitochondrial adenosine deaminases isozymes were 99% and 0.004% respectively.
3. In phyenylhydrazine-treated rabbits, total adenosine deaminase activity was maximal on 4th day, about 435 units, which is about 2.1 fold higher than that of normal value.
4. The increase in total adenosine deaminase activity during the induction of retiealocytosis appeared to ba due primarily to the 3.5 fold increase in the activity of the mitochondrial isozyme (KS = 0.0018 ; K_(d) = 0.0011 ; t1/2 = 630hr).
5. The activity of the cytosolic isozyme in pheny hydrazine-treated rabbit red blood cells increased to about 2.07 times than that of normal value (K_(s)= 4.055 ; K_(d)=0.0093 ; t1/2= 74.5hr).
6. The rate constants following recovery from phenylhydrazine treatment were ; K_(s)=0.0204 ; K_(d) = 0.0055 ; t1/2 =12.6hr, for the mitochondrial isozyme and K_(s) = 6.453 ; K_(d)=0.030 ; t1/2 =22.6hr. for the cytosolic isozyme.
